again after second time could be S+T+OVER which is a real word but that doesn't appear to be the name of a prayer (or a notable person known for praying), as far as i can tell

Conversely, I was thinking about S+T+RAIN (as in a piece of music) which is a very tenuous def of "prayer", but rain != again as far as I can tell

If "after second" could do double duty, we'd have "again after second [letter]" = AIN, after S (and then) T, yielding SAINT, one who prays. But it can't, that I know of.
Oh, oops, it's also too few letters.
