i wonder if the puzzle is any more interesting if operators are only allowed on one side of the equal sign
 
personally, I think the puzzle was somewhat 3/10 in terms of difficulty

the limit of 3 digits was to preclude 10101+0 = 10101 which would have been preposterously uninteresting
 
and, with leading zeroes... 10101 = 010101

"Two tablets" could refer to a diptych (but I can't find a 9-letter word for that), or to the ten commandments, for which we have DECALOGUE. If the definition were "two tablets", "take" would be awkward as filler; I don't know how to interpret it. I sort of doubt this is a fruitful direction.
I also considered that the definition might be "take two tablets", which could be ALLEVIATE or NARCOTIZE, etc.; the problem there is that all of the "synonyms" I can find of the right length are really too specific to mean "take two pills" generally. Something meaning ~ "medicate" would be better.
 
Or it's an &lit. for something like overdose.

The C4 hasn't been discussed much. The only serious discussion was our short braistorming on Wednesday. We'ev put out several possible defs, but not much wordplay, so looking at hint (2) might be useful.

oh, after our discussion I also found HEXAMINE, a substance used for fuel and sold in tablets
I tried to fit HEXAMINES. but I don't think "tablets" is a good definition for that anyway
