The tilde ("~") is shorthand for your home directory so you don't have to type the full path.
Context might make a different for the next part. Where in nginx are you seeing it used?
If it's being used in a regex statement, the "^" character means "the beginning of the string", so "^/users" might be a regex filter looking for string that begin with "/users".
Also, in some places, nginx uses ~ to indicate a case-sensitive filter.
