pacoverflowI used the Nero CD-DVD Speed program to scan the burn quality of a 4.7 GB DVD+R disc. It reported significant errors (PIF errors) at the 2.5 GB mark of the disc. On the underside of the disc, I see a small scratch about 13mm from the outer edge of the disc. Would that correspond to the 2.5 GB ...

The CLI does have the advantage of being easier to use remotely, if necessary.
The most important distinction is: "End Process" in Task Manager does exactly the same thing as taskkill. It uses the TerminateProcess API call (AFAIK)
"End Task" just sends a WM_CLOSE window message (or CTRL_CLOSE_EVENT if you target a console-mode program).

@user19679 There are edge cases where one or the other won't work. That's typically when you have something like a long I/O queue (so you can't even open the task manager or cmd programs), or if some driver has stalled the kernel (graphics drivers like to do that, so much that Microsoft moved most of it to userspace Vista and implemented TDR to auto-reload it if it stalls for a few seconds).
But in the general case, there isn't that much difference. Use whatever you're more comfortable with.
Oh, and there's no standard cmd equivalent for "End Task" (the 'nice' way)
You can visualise WM_CLOSE ("End Task") as politely asking the program to close, and TerminateProcess ("End Process") as shooting it and ripping its body apart.

When you politely ask, the program can save data and safely close all files it has open. It can maintain any structures in those files. It can prompt you to save files. (if it takes too long you'll get a popup asking you to "End Now" -- that does the same as "End Process" and kills it immediately with possible data loss)
This assumes the program handles the exit request properly, of course, but that's a necessary assumption.
If you kill the process, then it has no chance to do any of those things. It can die in the middle of an operation. The operating system will step in and make sure some things are consistent - it'll free any RAM the program was using, close file handles, etc., but if the program was, say, in the middle of writing a file that file's structure might now be corrupt.
Some programs, like database engines, will keep what's known as a "journal" where they write all intended actions before actually making changes to the real DB so if they crash unexpectedly they can replay the journal to maintain consistency in the real DB. Some limited error recovery, though often some (newly-added) data will still be lost, it tries to preserve the rest of the file.
