When you hover on "test" folder, it still shows blank popup :) And OP's question was Is there a way to use conditional formatting so that the pop-up only appears for rows that have text in the description column?

I am able to see the blank popup (in diamond shape) in the middle of Name and Modified columns, when you hover on "test" folder. Check the gif file you attached to your answer. Maybe you should check it again.

Maybe you didn't notice it because it is in very small size and white background
